Optogenetics
Optogenetics continues to supply neuroscience with ever more efficient tools for light mediated neuronal control and we are expanding our viral service to make these readily available to the community. We now offer AAV9 for 2 more constructs from Christopher Harvey’s lab. These plasmids localize opsins to the soma to avoid off target photostimulation of axons and dendrites on neighboring neurons. We have also packaged hChR2(H134R) plasmids from Karl Deisseroth’s lab, which includes pAAV-hSyn Con/Fon hChR2(H134R)-EYFP that is both Cre and Flp dependent.

Control AAV
Control AAV allow researchers to not only interpret their results, but also confirm expression in target cells. While the various AAV serotypes can be used to achieve different patterns of expression, researchers often need to determine these transduction patterns empirically for their own experiments. To this end, we offer specific control AAV in smaller aliquots of 20 ul for serotype testing to help you pick the best AAV for your experiment. The Addgene viral service has now expanded to offer many of our control plasmids in new serotypes.

Biosensor AAV
Calcium sensors
Tracking calcium dynamics that govern such activities as neurotransmitter release is vital in studying neural networks. We have expanded our calcium sensor collection to include our first Flp-dependent GCaMP, AAV8-105714. Several new options are also available for the calcium sensor jGCaMP7 from Janelia, including three jGCaMP7 plasmids now packaged in AAVrg. jGCaMP7s, the variant with the most sensitive response to action potentials, is now offered under control of the CAG promoter for ubiquitous expression. Neurotransmitter sensors: Dopamine and norepinephrine
Neuroscience research relies on accurate monitoring of neurotransmitters and we are excited to offer AAV for the newest GRAB sensors from the Li Lab. These GPCR based sensors provide the spatiotemporal resolution required for precise in vivo measurements of dopamine (113049, 113050) or norepinephrine (123308). The two dopamine sensors offer different levels of apparent dopamine affinity, with 113050 offering high apparent affinity, and 113049 offering medium apparent affinity.

Glutamate sensors
iGluSnFR is a useful tool from Loren Looger’s lab at Janelia for neuroscientists to measure glutamate levels. iGluSnFR.A184S, a variant with higher affinity to glutamate, has been packaged into AAVrg.
