A New DNA Format for the Plasmids You Already Use

Few lab routines are as commonplace — or as patience-testing — as DNA preps. For most labs, getting plasmid DNA still means streaking plates, growing an overnight culture, performing a miniprep, and checking concentration and quality. Researchers can spend a few days (or longer!) preparing each construct before the real experiment can begin.

Even after that time investment, a prep can fail for any number of reasons. Low yield, contamination, or poor purity can quickly derail your experiment. When that happens, you’re back at the bench, running it again, often days later than planned. To top it all off, variations from prep to prep can have you repeating all these steps in the future.

In a recent survey, researchers told us their in-house preps deliver unpredictable yields or variable purity, and only about one in five called their DNA preps trouble-free. When a timeline is tight, a prep that should have worked but didn’t can be a costly delay.

Transfection-Ready DNA, a new service from Addgene, saves you the prep (and the patience). It’s the same plasmids, from the same catalog you already know, just delivered ready to pipette instead of as bacterial stabs.

What is Transfection-Ready DNA?

Transfection-Ready DNA is a ready-to-use, high-purity DNA prep intended for mammalian cell transfection, though you can use it in a range of other molecular biology applications. Each order arrives as purified plasmid DNA, in a format you can add directly to your transfection mix of choice. Every prep includes:

  • DNA in Tris buffer (50 µg or 100 µg)
  • 1 µg/µL concentration
  • Low endotoxin levels (production process validated for <1.0 EU/µg)
  • Whole-plasmid sequence verification using next-generation sequencing (NGS)

Quality you can pipette with confidence

Of all the things researchers asked us for, quality mattered most. That’s why every Transfection-Ready DNA prep is fully sequence verified by NGS. Rather than relying on the spot checks that Sanger sequencing or a restriction digest provide, we confirm the whole plasmid. This way, you know exactly what you are using in your transfections, every time.

Another component of quality is purity. Endotoxins, which originate from the bacteria used to produce plasmid DNA, can significantly reduce transfection efficiency. Addgene has validated our production process to ensure consistently low endotoxin levels in every Transfection-Ready DNA prep (Figure 2).

What’s available now and what’s coming

We’re launching the transfection-ready format with more than 27,000 plasmids, from three of the most-requested categories of our collection:

  • CRISPR tools like Cas9, other Cas nucleases, and sequence-specific gRNAs
  • Imaging and reporter constructs such as fluorescent and luminescent reporters
  • Gene expression vectors for driving protein expression in mammalian cells

Each one is drawn from Addgene’s repository of 175,000+ constructs, so it’s traceable to the lab that deposited it and, for most plasmids, to the publication that describes it.

Choose the amount that’s right for your experiment, 50 µg or 100 µg, and place your order. We’ll ship your Transfection-Ready DNA within five business days of Material Transfer Agreement (MTA) approval.
